B. The steps of the air disinfectant neutralization identification test:

 

This method is suitable for the disinfection effect evaluation test of 20 m~30 m aerosol chamber, and the sampler is a six-stage sieve air impact sampler.

 

1. Preparation of bacterial suspension

 

Diluted with nutrient broth to obtain a bacterial suspension for the test with a concentration of 5×10CFU/mL~3×10^4CFU/mL

 

2. The neutralizer identification test is divided into three groups

 

Group 1: Draw 0.1 mL of the bacterial suspension for the test respectively, spread it evenly on two nutrient agar plates containing neutralizer, and count the viable bacteria culture. Observe whether the neutralizer has an inhibitory effect on the growth of the test bacteria.

 

Group 2: In accordance with the dosage of disinfectant required by the instructions, spray in a 20 m ~ 30 m bacterial aerosol chamber, immediately sample with a six-stage air sieve impact sampler containing a neutralizer nutrient agar plate (the air sampling volume is the same as the pre-sterilized sampling volume in the air disinfection identification test) for 10 minutes.

 

Draw 0.1 mL of the bacterial suspension for the test and apply it to six plates with disinfectant to count the viable bacteria (the average of the colony counts of the six plates is taken for the calculation of the results).

 

Group 3: Draw 0.1 mL of the bacterial suspension for the test respectively, spread it evenly on the surface of two ordinary nutrient agar plates, and count the viable bacteria.
